New hemodialyzers are routinely tested to establish their biocompatibility with the use of animal models that are more reliable than in vitro animal models used in the past. These in vivo assays include acute and chronic toxicity tests, as well as single or repeated dose bioassays. The development of such assays is expensive and time consuming, so a computationally aided prediction of their outcome would be useful. We have developed a new algorithm for predicting the chronic toxicity and blood compatibility of hemodialyzers. Three of the most important features of the algorithm are: 1) input is either the concentration of the extracellular fluid volume (ECFV) of a prototype of a new hemodialyzer or the volume exchange rate (VVR) of a prototype, 2) the output is predicted median lethal dose (LD(50)) values and blood compatibility (BC), and 3) the algorithm can be used in fast simulations to predict acute toxicity from the LD(50) values. The chronic toxicity is predicted accurately in most cases. We also predict blood compatibility for many dialysate chemicals. Importantly, the algorithm can be used in predicting acute and chronic toxicity in different organs of rats, as well as blood compatibility of hemodialyzers. (0.24--0.98)
